Metabolic Engineering of 2,3-Butanediol Biosynthesis in Saccharomyces cerevisiae

2,3-butanediol(2,3BD) has proven to be very important for a variety of chemical feedstocks and liquid fuels. For enhanced biotechnological production of 2,3BD, 2,3BD biosynthetic pathway has been widely explored in many organisms. In this study, for diversion of carbon flux to acetoin without accumulation of ethanol and acetate, Saccharomyces cerevisiae strain BY4741 lacking ALD6 and ADH1, which encodes acetaldehyde dehydrogenase and alcohol dehydrogenase, respectively, was generated and overexpressed PDC1 pyruvate decarboxylase. To balance NADH utilization and enhace the production of 2,3BD, GPD2 encoding glycerol-3-phosphate dehydrogenase is deleted and BDH1 encoding butanediol dehydrogenase is overexpressed. Finally these strains are fermentated about 3 days in YPD media (with 2% or 20% glucose) and bioproducts are analyzed by using HPLC and GC instruments.
