Many attempts have been made to decrease carbon dioxide which causes global warming. One desirable method to decrease carbon dioxide concentration is the use of carbon dioxide as a substrate for enzyme reactions for the production of industrial chemicals. Four-carbon metabolites are synthesized through carboxylation of three-carbon metabolites. Four-carbon dicarboxylic acid producing microorganisms are receiving attention to produce commodity chemicals from renewable feedstock. In this research, carbonic anhydrase(CA) for conversion from carbon dioxide to bicarbonate was used to fix carbon dioxide in the air and to convert to four-carbon metabolites in Escherichia coli. The carbonic anhydrase from marine micro-algae Dunaliella sp. was cloned into low copy expression plasmid pET21 and transformed to host strain E. coli BL21(DE3).
