The effect of Grapefrult Seed Extract by cosmetic preservatives

Grapefruit Seed Extract (GSE) have been well known as natural materials with its distinguished antimicrobial activities. In this study, antifungal and antimicrobial susceptibility of methyl paraben and propyl paraben, which are both synthetic and common cosmetic food grade preservatives, and also that of natural preservatives were tested, and by measuring preservative quality of lotion formulation, possibility of replacement of existing synthetic preservatives with a natural preservative was investigated. From the results of the test with liquid culture medium revealed that GSE had the highest antimicrobial effect among the test resulting materials having Minimum Inhibitory Concentration (MIC) value. Therefore, from the results of the test, it was confirmed that all natural materials had higher level of antimicrobial activity, and based on this test result, relevant cosmetics were made and the possibility of a preservative was tested accordingly. From the results of the test, it was confirmed that there were antimicrobial activities which were higher level than that of existing synthetic preservatives or similar to it in all formulations that contained each of GSE and the results from the test displayed that there were antimicrobial activities in a lively fashion at 0.03 %, 0.1 %, 0.1 % concentration, respectively. Also, from the results of the test of preservative quality according to mixture use of essential oil, it was unable to confirm the rising effects caused by mixture use, for it had displayed similar results to the antimicrobial activity of mono oil. In consequence, it was concluded that GSE could replace existing synthetic preservative effectively.

본 연구에서는 화장품에 사용되는 합성방부제인 methyl paraben, propyl paraben과 미생물에 항균성이 뛰어난 천연물질로 알려져 있는 천연방부제로서 Grapefruit Seed Extract의 향균력을 실험하고 로션제형에서의 방부력을 측정함으로써 화장품 제조분야에 사용되는 기존 합성방부제를 천연 방부제로의 대체 가능성을 알아보았다. Grapefruit Seed Extract에 대한 항균력 시험결과 disc paper method를 이용한 실험에서 Grapefruit Seed Extract 물질의 항균효과와 농도에 따라 항균활성이 증가하는 것을 확인하였다. 액체배지에 시험한 결과 최소 저해농도(MIC), 최소 살균농도(MBC)값의 실험결과 GSE는 우수한 항균효과가 나타나는 것으로 확인하였고, 시럼한 천연물질에서 높은 항균활성을 갖는 것을 확인하였고, 결과에 따라 화장품을 제조하여 방부제로써의 가능성을 시험하였다. Grapefruit Seed Extract를 각각 함유하는 모든 제형에서 기존의 합성방부제 보다 높거나 유사한 방부활성을 확인하였고 각각 0.03%, 0.1%, 0.1%에서 효과적으로 방부활성을 나타내었다. 이러한 결과를 토대로 GSE를 화장품 방부제에 적용할 경우, GSE를 함유한 제품에는 낮은 농도의 방부제만으로도 방부력 효과가 높을 것이며, 내용물의 안전성 또한 좋을 것으로 사료된다.
