Porcine epidemic diarrhea virus (PEDV) is a Coronavirus and a causative agent for diarrhea in pigs, particularly in neonates. The propagation of the PEDV in cell culture was developed in 1988. Porcine epidemic diarrhea disease vaccine was produced from Vero cells by using lively attenuated virus strain[1]. Several physiological or physical operation variables should be optimized for the virus vaccine production in cell culture. These operation variables include multiplicity of infection (MOI), infection, and harvest time of virus, temperature, pH, osmotic
pressure, stirring speed and gas supply rate in suspension culture and so forth. However, the most basic and important operation variables for virus production are MOI, and infection and harvest time. Therefore, we studied optimum MOI of various PEDV strain, such as PED-SM,
KPED-9, and DR-13. The optimal MOIs for three PEDV strain were all same as 0.1. In addition, the optimum infection (72 hr) and harvest time (24 hpi) for PED-SM production in static culture were determined. Other operation variables than MOI and infection and harvest time were also
optimized by following investigation.
