Phosphorylated chitooligosaccharides (P-COS) were prepared using H3PO4, P2O5, Et3PO4 and hexanol solvent system. The P-COS were characterized using various analytical techniques like FT-IR, TG-DTA, 13C NMR, 31P NMR, X-ray diffraction and solubility studies. The results
reveal that the phosphorylation occurs at C3 and C6 position of OH groups and C2 position of NH2 group. There is no decomposition was observed in pyranose ring in P-COS due to heating and acidic condition, was confirmed by infrared spectroscopy. The X-Ray diffraction analysis
showed that P-COS was amorphous in nature. Further, the biocompatibility and alkaline phosphatase activity of P-COS were checked against osteosarcoma MG63 cell line at different concentration. As a resultant, no cytotoxicity was found. After 7 days of cell incubation under the
conventional method, the ALP activity was higher compared with the control group. This results suggests P-COS is biocompatible materials and in future P-COS could open up a number of promising pharmaceutical and clinical applications to mankind.
