To produce sugar from hot water fraction of Schizocytrium sp. for bio-ethanol production, hot water fractionation conditions were optimized by a response surface methodology (RSM) based on the 23 factorial central composite design. Optimum conditions were at 115.5℃ temperature,
46.7 min time, and 25% (w/w) solid loading. The model predicted that maximum oligomeric sugar content which was recovered by hot water fractionation under the above optimum conditions was 19.4%. Experimental results were in close agreement with model prediction with 16.6%. In the fermentability test of sugar which was recovered from hot water fractionated Schizocytrium sp., 11.8 g-EtOH/L of bio-ethanol was produced from 25.7 g/L of glucose by E. coli KO11. The maximum bio-ethanol production yield was 88%.
