Various fungal strains producing cellulase were isolated from soils of glasses, wood, bamboo. The cellulase production was juged by the relatively large clear zones around the colonies on agar plate medium containing carboxymethyl-cellulose(CMC), after flooding of the agar plates with Congo red. The activities of CMCase and filter paper cellulase(FPase) were analysed by incubating the enzyme reaction mixture containing the culture filtrates and CMC or Whatman filter
paper NO. 1 at 50℃. The microogranisms with relatively higher CMCase and FPase activities were selected and were grown on agar media containing CMC, avicel or barley straw to produce enzyme extracts for analysis. Reducing sugars released from filter paper, avicel, barley straw,
CMC, pectin, arabinoxylan and cellubiose was quantified using a miniaturized colorimetric assay based on 3,5-dinitrosalicylic acid. The results showed variation in enzyme activities were observed among fungal species, growth media and substrates.
