Improvement of M3 Alginate Lyase Activity by Homology Modeling-derived Mutations

Poly-guluronate specific lyase from Streptomyces sp. strain M3 from marine algae was cloned and expressed in E. coli BL21(DE3), previously. Partialy purified M3 alginate lyase was showed temperature stability at 50-60℃, whereas ALG-5 lyase which has 7 different aminoacid residues
showed temperature stability at 30-40℃. So, we carried out homology modeling and compared the energy level between M3, 1UAI and ALG-5 alginate lyases using Prosa 2003 and AutoDock Vina program for improvement of enzyme activity or stability by site directed mutagenesis.
Homology modeling-derived mutagenesis showed the improving effects on alginate degrading activity.
