A specific poly-MG alginate lyase was cloned and characterized from an airborne Pseudomonas fluorescence KS408 (previous work). A homology model of this lyase has been constructed using the X-ray structures of Sphingomonas A1-III [Lyase, EC: 4.2.2.3] as templates, using MODELLER 7v7. The substrate MMG block was docked into the model allowing
further analysis of the active site architecture by using AutoDock software. The energy level of lyase was generated through ProSa2003 software, and the lowest energy model was then assessed for stereochemical stability. The important residues were identified to generate mutant
engineered lyase with high performance ability for further use. Acknowledgement: This work was financially supported by the Ministry of Knowledge Economy(MKE) and Korea Industrial Technology Foundation (KOTEF) through the Human Resource Training Project for Strategic Technology.
