Inonotus obliquus is a black parasitic fungus that grows on living trunks of the mature birch. And Inonotus obliquus has been traditionally used for both medicinal and food purpose. Polysaccharide isolated from Inonotus obliquus (PFIO) are able to stimulate the immune system and to exert anti-tumor activity through the stimulation of host defense mechanism. Macrophages are activated through innate immunity and cell-mediated immunity. Activated macrophages produce nitric oxide (NO), reactive oxygen species (ROS), and inflammatory cytokine. In addition, macrophages are phagocytic acting in innate immunity and cell-mediated immunity. Immunostimulating activity of macrophage is induced by phosphorylation of MAPKs (ERK, JNK, p38). Also, NF-κB nuclear translocation occurs from the cytosol to the nucleus. This study was aimed to elucidate immunostimulating activity and signaling pathway of activated macrophage by polysaccharide isolated from fruiting body of Inonotus obliquus
