The mixing effect of hydrophilic glycerol in hydrophobic dimethyl carbonate in lipase-catalyzed biosynthesis of glycerol carbonate was investigated. The hydrophilic glycerol was not readily mixed with hydrophobic dimethyl carbonate and they formed two phases when Novozyme 435 was used as the biocatalyst. When the reaction mixture was examined by microscopy, Novozyme 435 was coated with glycerol, which inhibits the contact between enzyme and substrates. This problem could be overcome by employing silica gel as the glycerol reservoir or
addition of detergents. Glycerol carbonate was successfully synthesized with more than 90% conversion from dimethyl carbonate and glycerol with a molar ratio of 10 by using Novozyme 435-catalyzed transesterification at 70oC.
