Lycopene Production in a Fed-batch Culture of Glycerol with Adding Glucose and l-arabinose Using Metabolically Engineered Escherichia coli

The effect of carbon source on lycopene production by metabolically engineered Escherichia coli was investigated in a 7-l fermentor. Glycerol was selected as a main carbon source and its initial concentration was determined to be 10 g l–1 because of the highest productivity of lycopene. In the fed-batch culture of glycerol with an additional carbon source, the highest productivity of lycopene was observed at 10 g l–1 glucose or 7.5 g l–1 l-arabinose. In the fed-batch culture of glycerol with glucose, lycopene production increased due to the significant
increase of cell mass. In the l-arabinose supplemented culture, lycopene production increased due to the increase of cell mass and lycopene content. In fed-batch culture of glycerol with glucose and l-arabinose, the significant increased production of lycopene was obtained by the synergistic effect of the two sugars. In the fed-batch culture, the cell mass, lycopene concentration, lycopene content, and lycopene productivity were 42 g l–1, 1350 mg l–1, 32 mg g–1, and 40 mg l–1 h–1 after 34 h, respectively. They were approximately 3.7-, 7.1-, 1.9- and 11.7-fold higher than those in the culture without glucose and l-arabinose, respectively. This is the
highest reported concentration and productivity of lycopene.
