Bacterial cellulose (BC) is free from hemicellulose, pectin and lignin. Along with its conventional use as a raw material in clothing, pulp and paper industry it is appropriate to be use in speaker diaphragm, tourniquet, thickener for ink as well as in artificial skin and artificial blood vessels preparations. Hence, the study of its various physical properties is considered important. BC was produced in static conditions by G. hansenii PJK over the surface of agar plates containing different amounts of agar. The various parameters i.e., water holding capacity (WHC), thickness, FTIR and XRD of the prepared BC were determined. The study revealed that the BC produced over the agar plates gave the same XRD peaks as the BC produced in the control. The thicknesses of the BC sheets produced over the surface of agar plates were higher compared to the BC produced in the control. The FTIR spectra were in agreement with the high WHC in case of BC produced in the surface modified reactors contained 2.5, 3.0 and 3.5 % of agar compared to the control.
