Reactive oxygen species (ROS) are known to cause DNA damage, oxidative modification of protein and lipid peroxidation. Oxidative stress is caused by ROS and accelerated by exposure of environmental oxidant and UV irradiation. This oxidative stress is known to associate with
tissue damage, aging, inflammation, cancer and diabetes. The purpose of this study was to evaluate cytoprotective and anti-aging effects of Inonotus obliquus extract, which was extracted with water at 80oC, against hydrogen peroxide (H2O2)-induced cell damage in human dermal
fibroblast (HDF) in vitro and UV-B-exposed hairless mice in vivo via anti-oxidant activity. Inonotus obliquus extract scavenged intracellular ROS and thus prevented H2O2-induced apoptosis and cellular senescence. As a result, Inonotus obliquus extract was found to reduce not only cell death but also premature senescence by oxidative stress in HDF cells. In addition, Inonotus obliquus extract was found to reduce matrix metalloproteinases (MMP) -1 and -9 activity and increase collagen synthesis and inhibit phosphorylation of extracellular signal-regulated kinase, which is activated by stress response, by modulating cellular signaling pathways. Further, it was found that Inonotus obliquus extract decreased UV-B irradiation-induced wrinkle formation and skin thickening in hairless mice. Therefore, the results suggested that Inonotus obliquus extract might be a potential agent for the prevention and treatment of
skin aging.
