A capacitive biosensor was developed based on chronoamperometry. In this work, the immunoaffinity layer was prepared by physical adsorption of antibodies and the electric isolation by the sequential adsorption of proteins at the immunoaffinity layer was estimated by using the redox reaction of Fe(CN)6 3-/4- by using cyclic voltammetry and impedance spectroscopy. The feasibility of the capacitive biosensor was tested by using HRP as a model analyte. The detection
range was determined to be from 100 pg/ml to 1 mg/ml and the limit of detection was estimated to be less than 100 pg/ml. The applicability of the capacitive biosensor was demonstrated by detection of Creactive protein (CRP) in serum. For the CRP samples, the detection range was estimated to be from 10 pg/ml to 1 mg/ml. The capacitive signal saturation was observed at the CRP concentration of 1 mg/ml. From these results, the capacitive biosensor with an immunoaffinity layer prepared by physical adsorption to the electrode can be effectively
used for the detection of protein analyte at the concentration range between pictogram and microgram per ml.
