In previous studies, recombinant 30K protein from Bombyx mori (silkworm) had shown many favorable effects on animal cells such as anti-apoptotic effect, cell growth and glycosylation pattern of recombinant proteins produced by cells. In this study, it was assumed that the
beneficial effects of recombinant 30K protein on animal cells could come from the enhancement of cell’s mitochondrial activity. In the MTT assay, normalized with cell counting, HeLa cells treated with recombinant 30Kc19 protein showed dramatically the enhancement in the MTT reduction. To see whether this activity increase actually came from the enhancement of mitochondrial activity, in vitro mitochondrial complex I / III activity assay was performed. The analysis showed a noticeable increase in the mitochondrial activity when r30Kc19 was
added to isolated mitochondria. These results highly imply that mitochondrial activity enhancement is closely related with beneficial effects of 30K protein on animal cells.
