Development of hypothermic preservation solution for the human dermal fibroblast using protein hydrolysates

Stable cell preservation is an essential factor in the regenerative medicine for cell therapies and transplantation of biologic materials. In this study, we studied to provide more stable hypothermic preservation by protection of cell damage during the preservation at 4℃. The result of searching for key components that have excellent efficacy in hypothermic preservation of cells, we have identified the fact that the hypothermic preservation adding protein hydrolysates such as yeast hydrolysate is far superior to others. All protein hydrolysates that are derived from animal, plant and microbe sources have superior efficacy, especially the peptides which have molecular weights under 10 kDa have the best efficacy among the components of protein hydrolysate. The protein hydrolysates prevented the decrease of ATP level in the cells caused by hypothermic environment and they inhibited the generation of ROS. Adding antioxidants and control agents of osmotic pressure were showed to have more superior efficacy in hypothermic preservation. Finally, KUL261 solution (DMEM/F12 1:1 medium, yeastolate 1%, α-tocopherol 100 μM, dextran 2.5%), the preservation solution developed in this study, showed the best efficacy in both cell viability and cell growth more than other conventional preservation solutions. In conclusion, the improved hypothermic preservation solution that contains the protein hydrolysates as a key component provide the best preservation efficacy. It provides better efficacy than other preservation solutions and will contribute to both the development of regenerative medicine and global commercialization in this therapeutic field.

최근 생명공학 기술과 의료수준의 향상으로 살아있는 세포를 이용하여 다양한 질병의 치료에 적용하는 세포치료 연구가 활발히 이루어지고 있다(1-3). 이에 따라 동물세포 의 이용성은 기존의 세포 생산물 이용 분야에서부터 세포 자체를 직접 이용하는 분야까지 그 범위가 확대되고 있다. 세포를 이용한 모든 치료 및 공정은 세포가 살아있어야 하 며 세포의 고유 기능이 유지되어 있는 상태여야 하기 때문 에 세포의 생존을 유지시키는 세포 보존기술은 세포를 이용 하는 모든 분야에서 매우 중요한 요소 기술이다. 세포는 보존과정에서 온도가 감소함에 따라 수많은 스트 레스를 받게 된다. 산화체계의 파괴로 활성산소가 생성되고, 이는 세포내에서 지질산화, DNA 및 RNA 손상, 세포골격 의 변화 등을 일으킨다. 또한 세포막의 구조나 유동성, 구성 분 등의 변화로 인해 막수용체가 활성화되고, apoptosis를 일으킬 수 있는 일련의 과정이 진행된다. 세포막의 Na+-K+ 펌프와 Ca2+ 이온 채널이 멈추면서 세포의 이온 균형이 무너지고, 이는 세포내 칼슘농도의 증가, 삼투압 차이 발생, 세포의 붕괴로 이어 진다(4-7). 저온보존에서의 세포 손상은 추가적인 스트레스 관련 반응을 유발할 수 있다. 온도가 낮아지면서 발생하는 대부 분의 반응은 세포에 손상을 일으키지만, 저온에서는 일부 세포의 생존반응이 활성화되기도 한다. 이는 세포가 스스로 스트레스를 줄이고 손상을 최소화하는 방향으로 반응하는 것을 말한다. 많은 경우에서 이러한 생존반응은 ATP와 같은 에너지를 더욱 빠르게 소비하기 때문에 결과적으로는 더 나쁜 상황이 되고, 결국에는 에너지 부족 등의 원인으로 세포사멸에 이르게 된다(8, 9). 세포나 조직의 저온보존에는 콜린스 (Collins)액, 유로콜 린스 (Euro-Collins)액, CelsiorⓇ액, HTK (CustodiolⓇ)액, UW (University of Wisconsin, ViaspanⓇ)액 등의 보존액 이 종래부터 이용되고 있으며, 현재 가장 널리 사용되고 있는 UW액은 다양한 장기 (organs)의 이식에서 주로 적용 되고 있다(10-15). 하지만 이러한 용액들도 실제 임상에서 는 장기를 24시간 이상 보존하기가 어려운 실정이다(11). 또한 장기 보존을 최적화하기 위해 설계된 용액이기 때문에 세포의 종류에 따라 보존능력이 상이하거나 그 효과가 미미 한 단점을 가진다. 따라서 최근에는 UW액에 항산화제를 비롯한 다양한 유효성분을 첨가하여 사용하는 등, 조직 및 세포 보존에 더욱 적합한 보존액이 개발되고 있다(16-18). 본 연구에서는 동물세포의 저온보존에서 우수한 보존능력 을 나타내는 유효성분을 탐색하고, 개선된 저온보존액을 개발 함으로서 보다 안정적인 세포 저온보존 기술을 제공하고자 하였다. 또한 동물세포 저온보존에 대한 향후 개발 방향을 제시하고자, 저온에 의한 세포손상 원인과 유효성분의 손상 보호효과를 평가하였다.
