A marine bacterium was isolated from seaweeds for its ability to degrade alginate. Analysis of 16S ribosomal DNA sequence and morphology revealed that the strain belongs to the genera Streptomyces. The alginate lyase gene of Streptomyces sp. ALG-M3 was cloned by using PCR with the specific primer designed from homologous nucleotide sequences. The alginate lyase gene consisted of 780bp and 259aa. The region from 1Met to 35Ala was expected to be a signal peptide by using SignalP 3.0 server. The consensus sequences of Nterminal region YXRXELREM, 152Gln, 154His and C-terminal region YFKAGXYXQ which is required for alginate lyase activity were conserved in the ALG-M3 alginate lyase gene. Purified ALG-M3 lyase was activated by 1 mM of CaCl2, MnCl2, CoCl2 and MgCl2, while 1 mM ZnCl2 and HgCl2 inhibited lyase activity completely. ALG-M3 appears to be poly-guluronate lyase degrading poly-G block specifically.
