Mussel adhesive proteins (MAPs) which can strongly affix to the wet surfaces, maintain its adhesive force and flexibility in harsh conditions. Protein crosslinking helps mussel withstand all hardships such as turbulence, humidity, salinity, and tides. By making bonds and tiying proteins together, they can alter their property to be hard. We successfully expressed mussel adhesive protein 151 in E.coli system and modified 151 mussel adhesive protein in vitro in order to make a DOPA containing 151 protein. To date, DOPA(3,4-dihydroxyphenylalanine) is believed as a key amino acid for adhesion. In mussel, DOPA covalently crosslink itself (auto-cross-linking) or form a multiple metal mediated ligands. In this study we tried to crosslink mussel adhesive protein and check the hardness of protein with water resistance effects. There are three types of cross-linking agents such as enzyme (tyrosinase), metal ions and chemical cross linker. We added a little amounts of tyrosinase for the auto-cross linker, sodium periodate(NaIO4) as a metal mediate crosslinker and glutaraldehyde which reacts predominantly with amino groups.
