Disposable Bioreactor Operation for the Production of hCTLA4Ig in Transgenic Rice Suspension Cultures

A disposable bioreactor based on wave-induced agitation has been for the production of various biopharmaceuticals by plant cell cultures.1) The advantages of disposable bioreactor are low cost, simple operation, simple scale-up and unnecessity of cleaning or sterilization process.2) Using a 20-L disposable membrane bag bioreactor, hCTLA4Ig could produced by transgenic Oryza sativa cultures. Maximum dry cell weight and specific growth rate (μ) in a 5-L stirred
tank bioreactor (STR type) control culture was 9.2 g/L and 0.19/day, respectively. Those value in a 20-L disposable bioreactor was 8.6 g/L and 0.13/day, respectively. Therefore, cell growth in the disposable bioreactor was comparable to that in STR. Maximum hCTLA4Ig production level reached 3.5 mg/L in the disposable bioreactor at day 20, which was 1.5-fold higher than
that in 5-L STR. In conclusion, we confirmed the possibility of using disposable bioreactor for the scale-up of transgenic plant cell cultures and the production of plant-made biopharmaceuticals.
