Enhanced Production of hCTLA4Ig with Protease Inhibitor Cocktail in Transgenic Rice Cell Suspension Cultures

Transgenic rice cell suspension cultures were utilized to produce hCTLA4Ig using α-amylase (RAmy3D) expression system. Stability of hCTLA4Ig was studied after the secretion of hCTLA4Ig into media. Stability of hCTLA4Ig was affected by various proteases that could cleave target protein in media during the production stage. The proteolytic degradation of hCTLA4Ig by proteases could cause post-secretion instability and the loss of productivity.1) The addition
of protease inhibitors into media could be an effective solution.2),3) In this study, the effects of the addition of Protease Inhibitor Cocktail (1, 3, 5, 7, 9% (v/v)) on the stability of hCTLA4Ig were investigated to increase the final level of hCTLA4Ig. It was found that the level of hCTLA4Ig was increased by adding Protease Inhibitor Cocktail compared with the value obtained without Protease Inhibitor Cocktail.
