Development of Bioprocessing for Vero Cell Derived JE Vaccine

The vaccine of Japanese Encephalitis Virus is produced in the brain of mice or fertilized eggs currently. However the conventional processes have many problems, such as the high cost of production and the controversy over bioethics for sacrificing animals. These problems could be resolved by the production of vaccine cultivated in mammalian cells. In this work, host cell growth kinetics and optimum conditions of Vero cell culture has been investigated. Cell growth kinetics showed that the maximum cell density with a serum free medium was about 1.5 times higher than that with serum containing medium (MEM-α with 5% FBS) but the cell growth was slower. The maximum density of Vero cells, 2 X 106 cells/ml could be obtained. The kinetics of nutrients consumption, metabolites secretion and virus proliferation will be presented.
