Eschscholtzia californica has been known as one of a traditional medicinal plant1) and shown to contain benzo[c]phenanthridine alkaloid used in the treatment of dental plaque and gingivitis.2) Among culture medium composition, nitrogen which exists in inorganic form, such as nitrate or ammonium in culture medium strongly influences on cell growth and secondary metabolites production. In this study, the effect of nitrogen source on cell growth and benzo[c]phenanthridine alkaloids production in suspension culture of Eschscholtzia californica were investigated. Experiments were conducted by modifying NH4 +/NO3 - ratio and total amount of initial nitrogen concentration in Murashige & Skoog (MS) medium. The increase of NH4 +/NO3 - ratio at total nitrogen concentration of 60mM caused decrease on cell growth and alkaloids production.
Total amount of initial nitrogen concentration also had influence on cell growth and benzo[c]phenanthridine alkaloids production. From this result, we found that ammonium inhibited cell growth and benzo[c]phenanthridine alkaloids production and then we conducted further study to verify ammonium function.
