Enhanced Production of Avermectin B1a by Medium Optimization and Random Mutation

Streptomyces avermitilis is produced by Avermectin which a secondary metabolite with powerful anthelmintic and insecticidal activities, thus it is used as an efficient agent in the field of agriculture and animal health. Avermectin is produced into eight derivatives which are A1a, A2a, B1a, B2a, A1b, A2b, B1b and B2b. We used to random mutation system to develop a strain that has high yield of avermectin B1a as a target, because avermectin B1a is the most
effective anthelmintic and insecticidal activities. Mutant strain was obtained via random mutation with NTG(3mg/mL,60minite) treatment. This mutant strain shows higher avermectin production (1.62mg/L) than wild type. For further improvement of avermectin production from mutant
strain, optimization of MF medium which is current avermectin production medium was studied. When fructose was used as a carbon sources, avermectin B1a was produced 12.7mg/L (7.8 times higher than MF midium) and when soytone peptone was used as a nitrogen source, avermectin B1a was produced 305.9mg/L (41.3 times higher than MF medium). We are proceeding more detail medium optimization for higher avermectin production and additional results will be discussed.
