Optimization of indigo production conditions by recombinant E. coli harboring fmo gene using response surface methodology

Response surface methodology (RSM) was very potential method for the optimization of medium composition and process factor. The optimization of medium components (tryptophan, yeast extract and sodium chloride) and culture conditions (pH and temperature) for production of indigo by recombinant E. coli harboring fmo gene was carried out using RSM based on the 23 and 22 central composite designs, respectively. The optimal combinations of medium constituents and culture conditions were determined as tryptophan 2.36 g/L, yeast extract 4.46 g/L, sodium chloride 11.40 g/L, temperature 30 and pH 7.0 respectively. When medium ℃ constituents were
optimized by RSM, the model predicted that the maximum indigo production which can be obtained was 781.3 mg/L, and experimental result was obtained as 785.9 mg/L. And then, optimization of culture conditions using RSM predicted enhanced indigo production as 852.7 mg/L. The maximum indigo production obtained experimentally was found to be 870 mg/L. These results were in close agreement with the model prediction.
