Chitin oligosaccharides (NA-COS) were prepared from hydrolysis of crab chitin in 12N hydrochloride acid for 7 hours at 40 0C, determined its chemical structure using FT-IR, NMR spectrometry and molecular weight (Mw) by TOF-Mass spectrometry, and their scavenging activities were investigated against 1,1-diphenyl-2-picrylhydrazyl (DPPH), hydroxyl, carbon-centered radicals using electron spin resonance (ESR) spin-trapping technique. Hydroxyl and carbon-centered radicals were generated from hypoxanthine-xanthine oxidase reaction, hydrogen peroxide-ferrous sulfate (Fenton reaction) and azo compound 2,2-azobis-(2-amidinopropane)-hydrochloride (AAPH), respectively. The results revealed that the radical scavenging activity of NA-COS increased with a dose-dependent manner and they also have reducing power. In addition, direct radical scavenging effect was determined in human fibro-sarcoma cell (HT1080) using the 2’,7’-dichlorfluorescein-diacetate (DCFH-DA) fluorescence probe. NA-COS with Mw 229.21- 593.12 Da exhibited cellular radical scavenging activity assessed by the increase DCF fluorescence intensity with incubation time.
