Oxidation of NAD(P)H for cofactor regeneration by mediator-free electrochemical method

Many oxidoreductases have been used to synthesize high value added compounds for pharmaceuticals, food additives, pesticide, and so on. Pyridine cofactors, which are NAD(H) and NADP(H), are essential components for oxidoreductase reaction. However, cofactors have to be regenerated for economically efficient enzyme process because it is too expensive to
stoichiometric use. There are many methods to regenerate cofactor like enzymatic, chemical, electrochemical, and photochemical method. Among all these methods, the electrochemical method is promising because the reaction is simple and enzyme for the cofactor regeneration is not required. Moreover, by-product is not produced in this system. In this study, cofactor-related enzyme reactions were performed. Cofactors were regenerated by electrochemical method. We
made a new electrode material for mediator-free cofactor regeneration. Oxidation of NAD(P)H was performed and it was applied to real enzyme reactions. Details will be presented.
