In this study, we describe a parallel generation and screening of ω-transaminases in a cell-free protein synthesis system. Open reading frames of 15 different ω -transaminases were simultaneously expressed in a 96-well plate. Screening of relevant substrates against the expressed enzymes was subsequently conducted in the same plate by colorimetric measurement of the changes in the concentration of an amine donor. Expression of 15 ω-transaminases and the subsequent screening of their substrate specificity against 12 keto acids was completed in 2 hours. Presented results demonstrate the high potential for the techniques of cell-free protein synthesis to be used a powerful tool for screening novel enzymes or
substrates.
