Ferritin is a globular protein consisting of 24 subunits to form a hollow shell and can store iron in the cavity.1) The iron storage protein is electrochemically active because of the reduction and oxidation of iron ions (Fe(II)/Fe(III)) in the protein shell. In addition, it was found that the naturally existing iron core could be readily extracted and replaced with a variety of electroactive materials. This finding enables ferritin to be applied to electrochemical applications such as biofuel cells.2) In the present work, ferritin was first immobilized to a gold electrode based on self-assembled monolayers. The immobilized ferritin was reconstituted through electrochemical removal of the iron core followed by chemical/electrochemical insertion of other metals into the cavity. Ferritins immobilized on the electrode were characterized by cyclic voltammetry. Applicability of the reconstituted ferritins to biofuel cells was examined.
