Effect of organic solvent and glutaraldehyde on the stability of permeabilized Ochrobactrum anthropi SY509 in nitrate removal

To increase the efficiency of denitrification by Ochrobactrum anthropi SY509, permeabilization of the cell membrane was performed. The stability of the permeabilized Ochrobactrum anthropi SY509 was 90 days in half life for denitrification, and should be increased for practical use. The activity of the growing cell was less than that of the permeabilized cell. The cell had viability
even though the cell was treated with the organic solvent for the permeabilization. The viability and stability of the cells depended on the type and concentration of organic solvent. Toluene was more effective than chloroform in decreasing the viability at the same concentration. But the concentration of toluene to make zero viability of the cells was 50 % and that concentration is too high to be applied for real operations. Crosslinking using glutaraldehyde was introduced to decrease the viability and increase the stability. Glutaraldehyde reacted with 40 % amine residue
on the surface of the cells during the crosslinking reaction, and the half-life of the permeabilized cells was increased from 90 days to 210 days.
