목적 : Capsular bag model에서 imatinib mesylate (IM, GleevecⓇ)가 수정체상피세포의 섬유아세포로의 전환분화억제에 미치는 영향을 알아보았다. 대상과 방법 : 돼지수정체상피세포를 여러 농도의 IM로 3분간 노출시켜 capsular bag model에 1주일간 배양한 후 전환분화의 정도를 섬유아세포들이 후낭을 덮는 면적과 α-smooth muscle actin (α-SMA) 양성 세포수를 통하여 비교하였고, 세포증식 정도를 BrdU면역염색으로 확인하였다. 또한, TGF-β를 첨가하여 전환분화가 촉진되는 지와 IM에 의해 TGF-β의 영향이 억제되는 지를 관찰하였다. 결과 : 섬유아세포들이 후낭을 덮은 면적은 30 &micro;M의 IM에서 유의하게 감소하였고(p<0.05), α-SMA양성 세포의 수와 BrdU양성 증식세포가 노출시킨 IM 농도에 비례하여 감소하였다. 첨가한 TGF-β 농도에 비례하여 후낭을 덮는 면적, α- SMA발현양성세포수 및 세포증식정도가 증가하였으며, IM의 농도에 비례하여 TGF-β의 효과가 억제되었다. 결론 : Capsular bag model에서 IM는 수정체상피세포의 섬유아세포로의 전환분화를 억제하였으며, TGF-β의 전환분화에 대한 효과도 억제함을 알 수 있었다.


Purpose: To investigate whether Imatinib mesylate (IM) could inhibit the transdifferentiation of the lens epithelial cells (LECs) into fibroblasts using the capsular bag model. Methods: In the capsular bag model, LECs were cultured by exposure to IM at various concentrations for 3 min. The effect of IM was analysed by observing the covering area, numbers of α-SMA positive cell, and BrdU incorporated proliferating cells. The same analysis was performed in the culture of LECs with TGF-β. Results: The covering area was significantly decreased by the treatment of 30 &micro;M IM, and the positive cells for α-SMA and BrdU were also decreased by IM treatment in a dose dependent manner. In addition, increasing TGF-β concentration accelerated transdifferentiation, but suppressed the acceleration of the transdifferentiation induced by TGF-β. Conclusions: In the capsular bag model, IM effectively inhibited not only the transdifferentiation of LECs into fibroblasts but also the TGF-β induced acceleration of the transdifferentiation.
