인체의 항상성은 세포의 재생, 분화 그리고 사망의 균형 속에 유지된다. Dexamethasone은 중간엽줄기세 포의 분화유도제로 오랫동안 사용되어 왔음에도 불구하고 이 세포의 증식과 세포자멸사에 대한 연구는 빈약한 실 정이다. 중간엽줄기세포의 증식, 분화, 그리고 자멸사 사이의 균형에 미치는 dexamethasone의 영향을 알아보고자 세포를 dexamethasone을 첨가한 군과 첨가하지 않은 군으로 나누어 3주간 배양하였다. 이 기간 동안 매주마다 세포수, DNA양, 분열세포수 자멸사세포수 그리고 alkaline phosphatase 검사를 하였다. 초기에는 dexamethasone을 처리한 군에서 DNA와 세포수가 낮았으나 3주째는 역전되었다. 세포분열은 두 군에 서 차이를 보이지 않았음에도 불구하고 세포자멸사는 dexamethasone을 첨가한 군에서 유의하게 낮았다. 결론적으로 중간엽줄기세포의 장기적인 고밀도배양에 있어서 dexamethasone은 세포자멸사를 억제하여 세포증 식에 영향을 미친다고 할 수 있다.

Dexamethasone has been used long time in field of MSCs research as differentiation inducer while, in contrast, there has been little work on the effect of dexamethasone on apoptosis and proliferation of MSCs. To determine the effect of dexamethasone on apoptosis and proliferation, MSCs were cultivated with (Dex) or without (Con) dexamethasone for 3 weeks. During this period weekly cell count, DNA assay, mitosis, apoptosis as well as alkaline phosphatase assay observation were recorded. DNA and cell number of Dex group was lower than Con group in early period but exceed at 3th week. There is no significant difference in mitosis between both groups whereas apoptosis frequency in Dex group was lower than that of in Con group. These results indicate that dexamethasone influences cell proliferation of MSCs in long term confluent culture by suppression of apoptosis.
